Identification of a new DNA polymerase activity in human KB cells.

We have isolated and partially characterized a new DNA polymerase activity from the purified nuclear fraction of cultured human KB cells. The physical and chemical properties of this enzyme which we have designated DNA polymerase N3 define it as a unique entity that can be distinguished from the several classes of eukaryotic D-DNA and R-DNA polymerase activity that have been recognized to date.     

A comparative analysis of the proteins from a cell culture and from field plants of the ecdysteroid producer Serratula coronata L.]

Differences in the composition and amount of proteins synthesized in the cell culture and leaves of field plants Serratula coronata have been shown. They proceed from differences in intensity of synthesis of secondary metabolites, ecdysteroids, whose content in the cell culture is considerably lower.     

Immunogenic properties of neomycin conjugates with macromolecular carriers]

To obtain diagnostic antibodies to neomycin, immunogenic properties of the neomycin conjugates with macromolecular carriers were studied. Bovine serum albumin and copolymers of N-vinylpyrrolidone with crotonic acid and N-hydroxyphthalimide ether of crotonic acid were used as carriers. It was shown that immunization of mice by the conjugates in combination with Freund's adjuvant resulted in production of neomycin antibodies, the titer being 1/80 to 1/130. When the antibiotic conjugates with the copolymers of N-vinylpyrrolidone were used and not the neomycin conjugates with the carrier of the protein nature, the neomycin antibodies were produced in the absence of Freund's full adjuvant. With the use of the isolated antibodies to neomycin a method for indirect solid phase enzyme immunoassay of neomycin was developed at the minimum detectable level of 25 ng/ml.     

Thermal stability of electron transport in PS II membranes and particles from the thermophilic cyanobacteria

Thermal stability of the ferricyanide (FC) and dichlorophenolindophenol (DCIP) reducing reactions was investigated in isolated membrane preparations and PS II particles with active water splitting system from the cyanobacterium Synechococcus elongatus. In a hypotonic medium, the thermostability was seen to be much higher for the DCIP than for the FC reduction reaction. After the addition of high concentrations of polyethylene glycol (Mwt = 4000) or sodium citrate to the medium, the FC reduction reaction appeared to be more temperature resistant. Data on the effects of temperature, DCMU and detergents on the electron transfer rate in PS II provide evidence suggesting that the different thermal stabilities of the two reactions are due to different physico-chemical properties of the electron donor sites to FC and DCIP. The data suggest that regions of contact between individual macromolecular complexes of the electron transport chain are the most labile sites of the photosynthetic apparatus. The role of the composition and properties of the intracellular medium on thermostability are emphasized.     

Expression pattern of the expanded noggin gene family in the planarian Schmidtea mediterranea

Noggin genes are mainly known as inhibitors of the Bone Morphogenetic Protein (BMP) signalling pathway. Noggin genes play an important role in various developmental processes such as axis formation and neural differentiation. In vertebrates, inhibition of the BMP pathway is usually carried out together with other inhibitory molecules: chordin and follistatin. Recently, it has been shown in planarians that the BMP pathway has a conserved function in the maintenance and re-establishment of the dorsoventral axis during homeostasis and regeneration. In an attempt to further characterize the BMP pathway in this model we have undertaken an in silico search of noggin genes in the genome of Schmidtea mediterranea. In contrast to other systems in which between one and four noggin genes have been reported, ten genes containing a noggin domain are present in S. mediterranea. These genes have been classified into two groups: noggin genes (two genes) and noggin-like genes (eight genes). Noggin-like genes are characterized by the presence of an insertion of 50–60 amino acids in the middle of the noggin domain. Here, we report the characterization of this expanded family of noggin genes in planarians as well as their expression patterns in both intact and regenerating animals. In situ hybridizations show that planarian noggin genes are expressed in a variety of cell types located in different regions of the planarian body.     

Stimulation of nucleic acid and protein synthesis in mungbean (Vigna radiata L.) seeds by uv irradiation.

The effect of a range of ultraviolet (uv) irradiation doses on nucleic acid and protein synthesis has been studied during seed germination and seedling growth in mungbean (Vigna radiata L). The treatment of seeds with low dose irradiation were stimulative for the synthesis of these molecules.
